Abstract: In this study, a novel rDNA based plasmid was developed for display of heterologous proteins on the cell surface of Yarrowia lipolytica using the C- terminal end of the glycosylphosphatidylinositol (GPI) anchored Y. lipolytica cell wall protein 1 (YlCWP1). mCherry was used as a model protein to assess the efficiency of the constructed plasmid. Y. lipolytica transformants harbouring the expression cassettes showed a purple colour phenotype on selective YNB-casamino plates as compared to control cells indicating that mCherry was displayed on the cells. Expression of mCherry on cells of Y. lipolytica was confirmed by both fluorescent microscopy and flow cytometry. Furthermore, SDS-PAGE analysis and matrix-assisted laser desorption/ionization (MALDI)-time-of (TOF)-mass spectrometry (MS) peptide mass fingerprinting (PMF) confirmed that the protein cleaved from the yeast cells using enterokinase was mCherry. Efficient cleavage of mCherry reported in this work offers an alternative purification method for displayed heterologous proteins on Y. lipolytica cells using the plasmid constructed in this study. The developed displaying system offers great potential for industrial production and purification of heterologous proteins at low cost.

Abstract: The sustainability of ICT for Development (ICT4D) initiatives, and indeed any development initiative, depends on many different factors that has been summarised in terms of financial, social, institutional, technological, and environmental sustainability. This complexity has led to researchers suggesting bricolage approaches that try to make do with the resources at hand, improvise and muddle through to develop local and contextual solutions. An important factor in this kind of approach is the role of relationships, particularly as evidenced in social networks of contact consisting of strong and weak ties, that has been called social capital and linking or bridging capital. The concept of social capital has been shown to influence many different processes in development. In the use of resources the capability approach refers to the influence of social capital on the conversion of commodities, technologies and resources by a person into situated use. The adoption of an innovation is also influenced by social capital, especially via the important role that is played by trust. In the design of development interventions, various types of theories of change have been articulated and the role of social capital in some of these theories is investigated. This paper aims to summarise and analyse the influence of social capital on development processes as seen from the different perspectives mentioned. In terms of development, a fundamental insight is that social capital plays a role in mediating development outcomes through embedded and autonomous social relations that can resolve social problems at macro and micro levels. Social capital also consists in crucial cross-level linkages that need to exist to enable top-down initiatives to meet bottom-up development.

Abstract: The present work aims to identify new promising plant sources, which could be exploited for their agrochemical properties. A total of 484 natural products from academic libraries were selected for screening against four fungal pathogens, five insects and two plants. On the basis of the hits founded and a literature survey, the flora of source countries (New Caledonia, French Guiana, Madagascar, Panama, South Africa and Greece) was analysed for plants containing the desired scaffolds. Lists of 1800 plant part samples were thus established. The plant parts collected generated 3600 extracts that are being evaluated.

Abstract: An investigation into the development of a wind energy industrial strategy for South Africa was undertaken by the Council for Scientific and Industrial Research (CSIR) and Risø-DTU of Denmark. This wind energy industrial strategy project aims to play a strategic role in paving the way for the gradual phasing in of wind energy in South Africa. The global and South African wind industries, energy markets and support mechanisms were critically reviewed where after a strategic analysis was undertaken to establish the economics of South African wind energy projects and scenarios for local manufacture were developed. It is beneficial to the South African economy that as much of the wind farm development costs are spent in South Africa as possible. Consequently, four scenarios for the localisation of wind-energy projects in South Africa have been proposed.
